Overview

Sermorelin is a synthetic analogue of endogenous Growth Hormone Releasing Hormone (GHRH), comprising its first 29 amino acids (GHRH 1-29 NH₂). It binds GHRH receptors on pituitary somatotrophs, stimulating pulsatile endogenous GH secretion and subsequent IGF-1 production. Unlike exogenous HGH, it preserves the natural pulsatility of GH release. Clinical studies in adults have used doses of 200–500 mcg. Previously FDA-approved as a diagnostic agent for GH deficiency (Geref); currently available only through compounding pharmacies. Synergistic with GHRP agents such as Ipamorelin in research settings. Evening timing is used in most study protocols due to alignment with natural GH pulsatility. Consult a licensed physician before use.

Research shows this combination achieves greater GH pulse amplitude than either compound alone, due to additive signaling at the pituitary somatotroph level.

Sermorelin is an FDA-approved synthetic mimic of GHRH (growth hormone releasing hormone), prescribed off-label for adults. Huberman discloses personal use: 200–400 mcg subcutaneously 3–5× per week, taken at night on an empty stomach. Derek (MPMD) tells Peter Attia that Sermorelin underperforms in practice: anecdotally people don't get the results they expect, and there is little literature clearly explaining why. In clinics, Tessamorelin + Ipamorelin has become the preferred GHRH/GHRP combo, largely replacing Sermorelin.
