Overview
Alpha-ketoglutarate (C5H6O5) is a central TCA cycle intermediate and epigenetic co-factor for TET enzymes and histone demethylases. A narrative review found AKG has pleiotropic metabolic, antioxidant, and immunoregulatory functions; AKG levels decline with age. Animal studies show longevity effects. Human clinical evidence remains limited. Available as calcium alpha-ketoglutarate. Investigational for healthspan extension and metabolic health. Generally well tolerated. Caution in active malignancies (TCA/epigenetic roles). No EFSA-authorized health claims.
