Histidine
Overview
Histidine (C6H9N3O2) is a semi-essential amino acid with a physiological pH-buffering imidazole ring. It is the precursor to histamine, carnosine (muscle pH buffer), and ergothionein. A 2025 meta-analysis found histidine-containing dipeptide supplementation (carnosine, anserine) significantly reduced depression scores with moderate certainty evidence. Mechanism: antioxidant, anti-inflammatory, metal chelation activity. Caution with histamine intolerance; avoid with MAO inhibitors. No EFSA-authorized health claims for isolated histidine.

Carnosine supplementation increases muscle carnosine levels fivefold and histidine twofold.
