Agmatine (C5H14N4) is the decarboxylation product of L-arginine functioning as an endogenous neuromodulator. Mechanisms: NMDA receptor antagonism, imidazoline receptor agonism (analgesic/antidepressant), NOS inhibition, and alpha2-adrenoceptor modulation. A narrative review described it as particularly promising for major depression with strong preclinical evidence. Human clinical evidence remains limited. Also a polyamine precursor (spermidine, spermine). NMDA antagonism may interact with psychoactive medications. Generally well tolerated in available trials. No EFSA-authorized health claims.
